Methodology

Useful For
Semen analysis is a critical part of an infertility work-up. Infertility is caused by the male in approximately 40% of cases. Evaluation of seminal fluid is relatively inexpensive and non-invasive when compared with other diagnostic procedures required for infertility work-up. It is therefore, one of the first procedures performed. This analysis is only to be used as a preliminary screen. If any parameter of this semen analysis is abnormal, the patient should be referred to a fertility center for the more labor intensive manual analysis.
Semen analysis consists of several parameters. Parameters determined by this procedure include Sperm count, volume, viscosity, pH, % motility and % normal morphology.
Performing Laboratory and Testing Schedule

Testing is performed on Monday thru Friday.
Must be scheduled in advance with Special Chemistry. Only available 0700 - 1400.
Schedule is subject to change based on holidays, instrumentation issues, and reagent availability.
Specimen Requirements
Specimen type: Semen
Specimen volume: Entire ejaculate
Minimum volume: 0.5 mL
Provide the patient with the instructions “Patient Instructions for Semen Collection” and verify that they have followed the instructions summarized below:
· 2-7 days abstinence from ejaculation prior to specimen collection
· Collect sample by masturbation only
· Lubricants, spermicides and other contaminants are not to be used
· Entire specimen must be collected into a clean container (preferably supplied by doctor’s office or lab)
· Clearly label specimen container with the patient name and date of birth or medical record number.
· Transport the specimen to the laboratory right after collection (if collected off-site)
· Keep the sample at ambient temperature, between 20 °C and 37 °C during transportation.
The semen sample should be tested within one hour of collection. Semen samples must be tested by the laboratory on a priority basis upon delivery - expedite to the testing area.
